Genetic risk score improved coronary prediction in diabetes trial
Researchers calculated a weighted genetic risk score from 204 variants across 160 loci in 5,360 white participants from the ACCORD trial and 1,931 white participants from the ORIGIN trial. The score was associated with major coronary artery disease events in both cohorts, independent of trial interventions. The association persisted after adjusting for classic cardiovascular risk predictors, though it was attenuated. Adding the genetic risk score to clinical predictors improved incident event risk classification, with a relative integrated discrimination improvement of 8 percent. The performance of this score was superior to scores based on smaller numbers of coronary artery disease loci available in previous years.
Combining genetic risk scores with clinical data may help doctors better stratify cardiovascular risk in people with type 2 diabetes. The benefit could grow as new coronary artery disease loci are discovered.
