Oral KRAS G12D inhibitor GFH375 shows responses in pancreatic cancer

The oral KRAS G12D inhibitor GFH375 produced a confirmed objective response rate of 35.1% in pancreatic ductal adenocarcinoma, according to phase 1 results in Nature Medicine. Median response duration reached 5.6 months. The trial enrolled 74 heavily treated patients with KRAS G12D-mutant advanced solid tumors, including 43 with pancreatic ductal adenocarcinoma. Another 16 participants had non-small cell lung cancer, and 10 had colorectal cancer. Dose-escalation across 22 patients showed no dose-limiting toxicities, establishing a recommended phase 2 dose of 600 mg once daily. One patient died from treatment-related septic shock.
