Polygenic Method Estimates Probabilities Across Three Psychiatric Disorders
The Psychiatric Genomics Consortium built DDx-PRS, a method that assigns probability estimates for schizophrenia, bipolar disorder, major depressive disorder, or control status from polygenic risk scores. The model trained on genome-wide association study data covering 41,917 to 173,140 cases per disorder (total n = 1,048,683). Tests on 11,460 held-out individuals showed the method was well calibrated and well powered. True diagnosis probabilities in the top deciles were considerably larger than prior baseline probabilities, according to results in Nature Genetics.
The method may help distinguish clinically overlapping psychiatric conditions using genetic data, though clinical utility remains to be demonstrated.
