Clocks trained to predict mortality or pace of aging produced the strongest and most consistent responses to pharmacological and lifestyle interventions. Study duration and participant characteristics influenced how biomarkers responded. Explainable clocks with multiple subscores provided more mechanistic detail than single-score clocks.
