Single-cell atlas of myeloma defines five malignant archetypes

A team analyzed single-cell transcriptomes from 341 patients with multiple myeloma and identified five recurrent malignant archetypes and a proliferation program linked to genomic features, treatment resistance and outcomes. The classification was validated in the CoMMpass cohort. The authors used a target-discovery pipeline to identify FCRL2, a plasma-cell surface protein, and showed that FCRL2-targeted CAR T cells killed myeloma cells in vitro and extended survival in mice. The work was published in Nature Genetics.

The classification may help doctors sort patients and choose treatments for multiple myeloma.
