Sixteen epigenetic aging clocks evaluated across fifty one human intervention studies
Researchers evaluated sixteen epigenetic aging clocks and ninety four DNA methylation biomarkers using TranslAGE, a database of fifty one human longitudinal interventional studies published in Nature Medicine. Clocks trained to predict mortality or pace of aging showed the strongest and most consistent responses across pharmacological and lifestyle interventions. Study duration, population characteristics, and multi score clock architectures significantly influenced biomarker responsiveness. The authors found that clocks with multiple subscores provide specificity and greater mechanistic insight into the responsiveness of interventions than single score clocks.
The findings can help design future clinical trials by guiding the choice of interventions and specific subsets of DNA methylation biomarkers to minimize multiple testing, study duration, study population and sample size.
