Study Compares 16 Epigenetic Clocks Across 51 Aging Intervention Trials
Researchers assembled TranslAGE, a database of 51 public and private longitudinal intervention studies, and calculated 16 different epigenetic clocks plus 94 other DNA methylation biomarkers for each study. The analysis, published in Nature Medicine, found that clocks trained to predict mortality or pace of aging showed the strongest and most consistent responses to pharmacological and lifestyle interventions. Study duration and population characteristics were key factors in determining biomarker responsiveness. Clocks with multiple subscores provided greater mechanistic insight than single-score clocks.
The findings can guide the design of future clinical trials by helping researchers select which epigenetic clocks and DNA methylation biomarkers are most likely to respond to aging interventions, potentially reducing study duration and sample size requirements.
