Werner helicase inhibitor RO7589831 produces partial responses in 7 of 66 MSI patients

RO7589831, a Werner syndrome helicase inhibitor, was given to 88 patients with advanced microsatellite instability solid tumors in a phase 1 trial reported in Nature Medicine. Seven of 66 efficacy-evaluable patients had confirmed partial responses, and the median response lasted 10.2+ months. Disease control rate was 74.2 percent. The trial did not identify a maximum tolerated dose. Three of 88 patients stopped treatment because of adverse events.

Patients with advanced mismatch repair-deficient cancers could gain a targeted option if later trials confirm the antitumor activity.
